Molecular Cloning (TSH) f3 Subunit of Quail Thyroid-Stimulating Hormone
نویسندگان
چکیده
Thyroid-stimulating hormone (TSH) fi subunit cDNA was obtained from Japanese quail (Coturnix japonica) by PCR and its nucleotide sequence was determined. The cDNA encodes a putative signal peptide and a mature protein consisting of 20 and 114 amino acids, respectively. The amino acid sequence of quail TSHf subunit shows homologies of 67-69% in mammalian species, 58% in amphibian and 43-49% in teleost fish. Comparison of the amino acid sequence with TSH$ subunits of other species reveals some differences in several regions responsible for its biological functions and characteristic features of the avian TSH/3 subunit, suggesting that the functional domains have diverged cooperatively between the hormone and its receptor during evolution.

Cloning of a partial cDNA for Japanese quail thyroid-stimulating hormone and effects of methimazole on the thyroid and reproductive axes.
The purposes of this study were to develop a probe for the detection of thyroid-stimulating hormone (TSH) beta subunit mRNA, to validate the usefulness of that probe in measuring TSH, and to use it to investigate the effects of thyroid suppression on TSH and the reproductive axis in Japanese quail. The objectives of experiment 1 were to isolate and characterize a partial cDNA for quail TSH and ...

Background and purpose: Measurement of thyroid stimulating hormone (TSH), usually through RIA and ELISA tests, is very useful for the diagnosis of thyroid disorders. Considering the structural similarity between alpha chain of TSH and the three glycoprotein hormones of luteinizing hormone, follicle stimulating hormone, and chorionic gonadotropin (CG), in this study, we aimed to examine the prod...
متن کاملA monoclonal thyroid-stimulating antibody.
The thyrotropin receptor, also known as the thyroid-stimulating hormone receptor (TSHR), is the primary antigen of Graves disease. Stimulating TSHR antibodies are the cause of thyroid overstimulation and were originally called long-acting thyroid stimulators due to their prolonged action. Here we report the successful cloning and characterization of a monoclonal antibody (MS-1) with TSHR-stimul...
متن کاملEffects of Thyroid Hormone (T4, T3 ), Bromocriptine And Triac on Inappropriate TSH Hypersecretion
Inappropriate TSH hypersecretion was diagnosed in a 34 years old woman. Earlier she had been treated by ablative therapy for hyperthyroidism. The present diagnosis was based on elevated basal serum TSH levels despite elevated serum free thyroid hormone level. Her TSH responses to TRH (peak value 240 mu/I) was high. Her albumin and prealbumin levels were normal. The serum TBG level, alpha subuni...
متن کاملNegative Regulation of the Thyrotropin Gene by Thyroid Hormone
Thyroid hormone (T3 and T4) is secreted from the thyroid gland, and is known to reduce the level of serum thyrotropin (thyroid-stimulating hormone, TSH) in the pituitary gland (Sarapura et al., 2002; Shupnik et al., 1989) (Fig. 1A). This is a typical example of negative feedback between the pituitary and endocrine organs, and is a key component of thyroid hormone homeostasis. TSH is one of the ...
